LM 12 OBJECTIVES & ACTIVITIES: IDENTIFICATION OF BACTERIAL UNKNOWNS-2
It is a standard part of most introductory microbiology labs to include an activity requiring students to apply concepts that have been learned in the course to identify unknown bacterial cultures. This is also an important activity in a clinical setting whereby a culture is sent to the lab for a culture and sensitivity. Typically the activity would assess your ability to perform the gram stain, describe cell morphology, grow the bacteria in pure culture so that colony morphology could be observed, make aseptic transfers, and conduct a series of selective and differential biochemical tests.
Unknown bacteria may be identified in the lab by microscopic analysis and completion of a series of biochemical assays, then using a dichotomous key (like the one shown here).

To complete this exercise, you will use your understanding of topics learned in the course, while still learning about the overall process of unknown bacterial identification. At the end of this unknown, you will write a report about your findings, and upload to Turnitin.

LM 12: Unknown Project Introduction-2
A patient presents to the clinic and is diagnosed with an infection. A nurse practitioner takes a sample of the infected area and sends the sample to the lab to be analyzed.
Preliminary gram stain on the sample indicates that it is a polymicrobial infection, having two unknown species, one gram positive, and one gram negative.
You will receive a mixed broth with the unknown patient number that contains both organisms. You must perform the necessary steps for isolation of both of these species and identify them both. Start by following the understood typical steps for isolation, subculture, and evaluation of pure cultures. Document each step of your process. During and after isolation and testing, you will use a dichotomous key flowchart to identify both of the unknowns in the sample. Additionally, you will determine appropriate antibiotics to use for this infection.
Summary of Procedures – How do you do this?
Each student will be given a mixed broth, and use the dilution streak plate technique to create pure colonies of their two organisms from the mixed broth culture. Students will use TSA, PEA, and MacConkey media.
Following incubation, the student will check to see if pure colonies of each organism have been obtained by observing colony morphology and by gram staining. TSA pure subculture plates will then be prepared of each organism. If colonies are still mixed, additional TSA plates may be streaked until each organism is isolated. The use of selective media greatly improves the chances of successful separation, but their availability is limited – use resources wisely!
Pure culture TSA plates of each organism must be observed for colony morphology. In addition, cultural characteristics on TSA and on any selective media used must be recorded. Additional media (MS and TSA/blood will also be available). Students may want to inoculate additional TSA streak plates. Purity of TSA subcultures should be evaluated using skills students have developed in lab. Students will then begin recording observations to IDENTIFY BOTH ORGANISMS and record results on Unknown Forms.
Each organism, once isolated and purified, will be tested using all necessary, available biochemical media, including a set of required biochemical tests for each organism. See posted flow charts for appropriate biochemical tests needed for identification. All tests needed to identify your organism must be recorded, included any on your forms that are mandatory tests. You MAY NOT inoculate all biochemical tests available in lab! Be critical and efficient in your media usage. Record all results that you test.
Each pure organism, will be subjected to Antibiotic Sensitivity testing using the Kirby Bauer method. Inoculation and interpretation will be done in class. Results must be recorded using the appropriate handout.
Each student must turn in a streaked TSA plate with isolated colonies. Streak plates MUST be labeled with Student Name, Class, Unknown number, and G+ or G-.
Once the identity of each organism has been obtained, a complete Unknown Report must be submitted, along with all report forms. Guidelines are in this lab module. You should thoroughly communicate your observations and results of isolation, pure culturing, biochemical testing, and KB tests. The Report is turned in online using a Turnitin link in this module.
What do you need?
The Form you will use to record your results and observations should be downloaded from the LM 12: Activities and Objectives page.
You will be required to turn in a copy of the Unknown Form by including it in your report. You may fill it in electronically, or by hand. If you fill it in by hand, you will need to scan it or take a picture of it when completed, and insert it into your unknown report. Make sure you refer to previous labs for the proper notations for each test. You will also include a brief note about a color or reaction you visibly see for each item, if applicable. Do not elaborate or go into details about your interpretation on the Form (instead, do so in the discussion of your Report).
Once all your observations are recorded properly, use the Flowchart found on the LM12: Activities and Objectives page to help you determine your unknown organisms identity. The Antibiotic Sensitivity Chart that is included in the third page of the document will help you evaluation your Kirby Bauer results.
