resealable snack size baggie
10 mL or 3/4 tablespoon extraction solution (100 mL or 1/2 cup of shampoo or dish soap, 15 g or 3/4 tablespoon table salt, and 950 mL or 4cups of water)
funnel
cheesecloth (2-3 layers, enough to cover funnel)
disposable cup (preferably transparent)
Isopropyl or ethyl alcohol (70%, ice cold)
wooden skewer/chopstick
collection container
ProcedureMash the strawberries in the baggie (remove air first). Try to not perforate your baggie, or the liquid will leak in subsequent steps.
Add 10 mL or 3/4 tablespoon of the extraction solution.
Continue mashing (again, remove air first). This step is called homogenization, as you make the mixture in the bag evenly mixed. Try not to make bubbles.
Pour the mixture through a funnel lined with cheesecloth. The solution will be thick, and you must be careful to not allow it to overflow.
Gently twist the cheesecloth until about 10 mL or approximately 3/4 tablespoon of the solution comes through.
Using a medicine dropper, carefully drizzle 10 mL or 3/4 tablespoon of ice-cold Isopropyl or ethyl alcohol down the side of the disposable cup. Try to not disturb the strawberry solution. The Isopropyl or ethyl alcohol will form a layer on top of the strawberry mixture, and it is at the interface of these two layers where the DNA will gradually precipitate.
Put a wooden chopstick or skewer into the center of the cup and slowly rotate the stick. DNA should “spool” (like thread) onto the wooden stick.
Transfer the DNA into either a collection container/ separate disposable cup.
If necessary, wait a few minutes and you may see that you can “spool” out more DNA from the interface of the two liquids. Add this DNA to the collection cup as well.
