This paper is based on what I did. My unknown sample was number 2.
I performed gram stain and it had purple grape like structures so that would be postive
I performed KOH procedure and obsereved there were no sticky strings that were pulled
i performed MAC which was inhibited ( no color change )
i performed blood plate which indiciated beta hemoloysis
I perfefirned the catalyse procedure to see if its streoptocci or staphlococcus and since it tested for staph
the unkown bacteria would be considred staphloccocus aures
I performed Kirby-Bauer Test
The test is used to determine a bacterium’s susceptibility to different antibiotics. Using a
sterile inoculating loop, the liquid culture to be used was prepared using the McFarland
Standards. A sterile inoculating loop was used to grab a decent amount of bacteria which was
then submerged and mixed into the liquid culture. This step was repeated until the desired
cloudiness was achieved. Using a p1000 pipette, 200 L of the liquid culture was pipetted onto
the surface of the Mueller-Hinton plate. Then, using a sterile spreader, the solution was spread
evenly throughout the plate. 3 antibiotic disks, chosen due to the Gram stain test, were placed
into the plate. A control disk was also added to the plate as a control for comparison. The plate
was then incubated at 37°C for 24 hrs.
